Consult the restriction map of the 20 -kilobase pair linear DNA fragment below to answer the following questions. (a) Draw a hypothetical DNA (i.e., agaorse) gel with the following four lanes: (1) molecular weight markers of increasing 5 kilobase pairs DNA standards (i.e., \( 5 \mathrm{kbp}, 10 \mathrm{kbp}, 15 \mathrm{kbp} \), and \( 20 \mathrm{kbp} \) ), (2) DNA fragment after EcoRI digestion, (3) DNA fragment after PstI digestion, and (4) DNA fragment after digestion with both EcoRI and PstI. (b) You subsequently perform a Southern blot. Specifically, you blot the DNA fragments onto nitrocellulose paper and probe the blot (i.e., the nitrocellulose paper) with the radioactive oligonucleotide labeled "probe," which is complementary to the indicated sequence and expose a piece of x-ray film to the blot. Indicate where you would expect to see bands on the film. Use the same four lanes that you had used in part (a).
